川贝母对哮喘小鼠Notch2的表达及炎症反应的影响 被引量:8

Effect of fritillariae cirrhosae bulbus on Notch2 and inflammatory response in asthmatic mice

摘要 目的探讨川贝母对哮喘小鼠肺组织Notch2的表达及炎症反应的影响。方法 36只BALB/c小鼠,雌雄不限,随机分为对照组、哮喘组和川贝母组,卵蛋白致敏的方法制备哮喘小鼠模型。取肺组织行HE染色;用乙酰甲胆碱行支气管激发试验,测定各组小鼠气道阻力的变化;免疫组织化学和Western blot方法检测各组小鼠肺组织Notch2的表达;ELISA方法检测各组小鼠血清中炎性因子IL-8、TNF-α、NF-kB的含量。结果哮喘组小鼠肺组织支气管周围炎性细胞浸润明显,川贝母组小鼠肺组织炎性细胞浸润减弱.而对照组无炎细胞浸润。川贝母能够降低哮喘小鼠的气道阻力,降低哮喘小鼠肺组织Notch2的表达,降低哮喘小鼠血清中炎性因子IL-8、TNF-α、NF-kB的含量(P<0.01)。结论川贝母降低哮喘小鼠气道阻力及肺内炎症,与下调Notch2的蛋白表达相关。 Objective To investigate the effect of Fritillariae cirrhosae bulbus(FCB)on Notch2 expression and inflammatory response in lung tissue of asthmatic mice.Methods 36 BALB/c mice(male and female)were randomly divided into control group,asthma group(ovalbumin sensitization)and Fritillariae cirrhosae bulbus group.Lung tissue was stained with HE,bronchial provocation test with methacholine was used to determine the changes of airway resistance in mice of each group.The expression of Notch2 in lung tissue was detected by immunohistochemistry and Western blot,and contents of inflammatory factors IL-8,TNF-a and NF-kB in serum were detected by ELISA.Results Inflammatory cell infiltration around bronchi was obvious in asthma group,but decreased in Fritillariae cirrhosae bulbus group,while no inflammatory cell infiltration was found in control group.Fritillariae cirrhosae bulbus downregulated the airway resistance,Notch2 expression level in lung tissue and the contents of inflammatory factors IL-8,TNF-a and NF-kB in serum in the asthmatic mice(P<0.01).Conclusion The downregulation of airway resistance and pulmonary inflammation factors by Fritillariae cirrhosae bulbus is related to downregulation of Notch2 protein expression in asthmatic mice.
