摘要
目的 :观察糖瘀平丸对链脲佐菌素 ( STZ)糖尿病大鼠肾组织糖化终产物 ( AGEs)的影响 ,探讨益气养阴活血法治疗糖尿病及慢性并发症的机制。方法 :对 STZ诱发的糖尿病大鼠分别给予糖瘀平丸高剂量 ( 8g/kg·d- 1 )、中剂量 ( 6g/kg· d- 1 )及低剂量 ( 4g/kg· d- 1 )灌胃 ,对正常对照组及糖尿病模型组分别给予等体积的蒸馏水灌胃 ,药物对照组给予水飞蓟素 ( 10 0 m g/kg· d- 1 )灌胃。处理 9周后 ,观察各组大鼠血糖、果糖胺及肾皮质 AGEs等指标的变化。结果 :糖瘀丸平能降低血糖 ,对早期糖化产物果糖胺及肾皮质 AGEs均有抑制作用。结论 :糖瘀平丸可降低血糖 ,抑制糖尿病大鼠蛋白非酶糖基化反应 ,提示其可能具有控制糖尿病发展 。
Object:To investigate the effect of tangyupin on inhibition of non-enzymatic glycation in kidney of strepozotocin(STZ)-induced diabetic rats. To explore the therapeutic mechanism of chronic diabetic complications by the pathogenesis of deficiency of both qi and yin,blood stasis and stagnation.Methods:The STZ-induced diabetic rats in groups of high and middle and low doses of tangyupin were perfused through mouth into the stomach respectively with 8,6 and 4 g·(kg·d) -1 tangyupin one time each day.Similar volume of distilled water was syringed through mouth into the stomach of rats in normal and model control groups.Silymarin(100mg·(kg·d) -1 ) was given by gastrogavage to the STZ-induced diabetic rats in medicinal control group.Rats were killed after 9 weeks of treatment.Fasting blood glucose (FBG),serum fructosamine (FMN) were measured. the fluorescence intensities of AGEs in renal cortex were also measured.Results:The levels of FBG in tangyupin groups were significantly lowered than those of untreated diabetic group.The early non-enzymatic glycation products-fructosamine and the advanced glycation end products in renal cortex were significantly inhibited in tangyupin groups.Conclusion:According to the pathogenesis of deficiency of qi-yin and blood stasis,the method of supplementing qi and nourshing yin and promoting blood circulation might decrease the levels of blood glucose,inhibit non-enzymatic glycation of protein,so as to slow down the advance of diabetes mellitus and prevent diabetic nephropathy.
