逆转录病毒介导的Flt-1M基因对大鼠胶质瘤的治疗作用

摘要 目的观察Flt-1M基因治疗对大鼠胶质瘤血管形成及肿瘤生长的影响。方法将转染了pLXSN-Flt-1M的产病毒细胞注入大鼠额叶胶质瘤瘤体内。随后处死动物,检测肿瘤的直径、微血管密度及凋亡细胞。结果肿瘤的微血管密度和直径都较对照组明显降低,而凋亡细胞则显著增多。结论Flt-1M基因治疗对大鼠胶质瘤具有显著的抗血管形成和抗肿瘤生长作用。 Objective To evaluate the therapeutic effects of Flt-1M gene mediated by retroviral vector on glioma in rats.Methods C6glioma cells were injected stereotacticly into the frontal lobes of cerebral cortexes to induce transplanted gliomas in the rats and5days later,the gliomas were treated by injection of virus-producing cells(VPC)transfected with pLXSN-Flt-1M into the gliomas.The rats were sacrificed20days after tumor implantation to determine the size,microvascular density(MVD)and apoptotic cells of the tumors.Results After treated with large numbers of VPC,the diameter and the MVD in the tumors significantly decreased and more apoptotic cells were observed within the tumors by TUNEL assay in VPC treatment group compared with the control group.Conclusion The Flt-1M gene mediated by the retroviral vector has significantly antiangiogenic and antitumor effects on the gliomas in the rats.[
