摘要
冰缘植物高山离子芥具有极强的抗寒能力,具葶离子芥为其同属近缘种,但抗寒性远不及高山离子芥。本试验以高山离子芥和具葶离子芥植株为材料,对这2种离子芥进行低温处理,并采用c DNA扩增片段长度多态性(c DNA-AFLP)技术对其进行耐冷相关基因筛选,共获得与耐冷相关的28条差异表达片段,其中7条来自具葶离子芥,21条来自高山离子芥,并对这28条差异表达的片段进行生物信息学分析。选取高山离子芥特有的5条差异表达片段(TDFs),采用Realtime RT-PCR进行低温诱导表达验证,结果表明确实是低温诱导的特异表达片段。本试验结果为高山离子芥特有耐冷基因的功能研究及其应用提供了依据。
Chorispora bungeana Fisch.et Mey is a kind of periglacial plant which exhibits cold tolerance,while Chorispora greigii Regel,a closely related species,shows weak cold tolerance.In this study,the gene expressions between the two kinds of Chorispora at low temperature were compared by c DNA amplified fragment length polymorphism technique.Twenty-eight transcript-derived fragments (TDFs) were identified,in which seven TDFs and 21 TDFs belong to C.greigii and C.bungeana,respectively.Realtime RT-PCR confirmed that five TDFs from C.bungeana were specifically expressed at low temperature.These results provided the basis for function analysis and utilization of cold related genes in C.bungeana Fisch.et Mey.
