大鼠血浆中10-羟基喜树碱含量测定方法

摘要 实验旨在建立大鼠血浆中10-羟基喜树碱含量的测定方法。通过高效液相色谱荧光检测HCPT和CPT的含量,建立了大鼠血浆中HCPT的浓度(X)与HCPT的峰面积和CPT的峰面积之比(Y)的线性回归方程以及回归系数。HCPT和CPT均可与血浆中的其他成分较好地分离,血药浓度在6.25~400 ng·mL-1范围内呈良好的线性关系,回归系数为0.9988。这种方法重现性好,操作简便,适用于血浓度检测。 This experiment aims at developing a method for determining 10-OH camptothecin content in rat plasma.Detecting the content of HCPT and CPT by using high performance liquid chromatography( HPLC) with fluorescence,this experiment establishes the linear regression equation and regression coefficientsabout the concentration of HCPT(X) and the peak area of HCPT ratio to the peak area of CPT( Y),and acquire regression coefficient in rat plasma. The result shows that HCPT and CPT in plasma can be better separated from other components,the concentration of plasma shows good linear relationship in the range of 6. 25 ~ 400 ng /mL and regression coefficient is 0. 9988. This method is easy to operate,has good reproducibility,and can be used to investigate the blood concentration.
