摘要
目的鉴定环肽化合物NW-G01对肿瘤细胞增殖与凋亡的调控作用。方法利用显微拍照法检测细胞的形态变化,克隆形成实验检测肿瘤细胞的克隆形成能力,MTT实验检测细胞增殖情况,流式细胞术测定细胞凋亡率。结果显微拍照法和MTT实验检测结果表明,NW-G01能抑制多种肿瘤细胞的增殖,其抑制效果具有时间依赖性和浓度依赖性。NW-G01对胃癌细胞SGC-7901的克隆形成能力具有显著抑制作用,并能降低细胞中S期细胞的比例,阻断细胞周期的进程。此外,流式细胞术检测结果表明,NW-G01能剂量依赖性地诱导SGC-7901细胞凋亡。结论环肽化合物NW-G01能抑制肿瘤细胞增殖,并诱导肿瘤细胞凋亡,在细胞水平具有显著的抗癌活性。
Objective To identify the effect of cyclic peptide compound NW-G01 on tumor cell proliferation and apoptosis. Methods Microphotography was used to detect cell morphological changes. MTT method was used to determine tumor cell proliferation,and the clonogenic survival assay was used to measure the effect of NW-G01 on tumor cell colony formation. Cell cycle was determined by measuring the percentage of Ed U positive cells and apoptosis was analyzed by flow cytometry. Results NW-G01 inhibited proliferation of various tumor cell lines in a dose-and time-dependent manner,and the IC50 ranged from 9 to 30 μmol/L after 72 h treatment. Moreover,NW-G01 significantly suppressed the colony formation of SGC-7901 cells and decreased the percentage of S phase cells. In addition,NW-G01 dose-dependently induced apoptosis of SGC-7901 cells. Conclusion NW-G01 significantly induces cell proliferation inhibition and apoptosis in tumor cells,indicating that NW-G01 is a promising compound for tumor treatment.
