摘要
目的 观察体外培养的大鼠第一磨牙牙乳头细胞的生物学特征。方法 大鼠第一磨牙牙乳头细胞培养 ,免疫组织化学染色、钙盐染色。结果 培养的大鼠牙乳头细胞表达Ⅲ型胶原、纤维黏连蛋白、碱性磷酸酶 ,当细胞培养于含 5mmol/Lβ-磷酸甘油钠和 5 0 μg/mlL -维生素C的培养液中时 ,细胞可形成矿化的胞外基质。结论 培养的大鼠牙乳头细胞保留体内的某些特征 。
Objective To observe the biological characteristics of cultured dental papilla cells in vitro from rat first molars. Methods Dental papilla cells cultured in vitro and stained by immunohistochemistry and by crystal of calcium. Results The cultured rat dental papilla cells expressed type Ⅲ collagen, fibronectin and alkaline phosphatase intracellularly. When incubated with 50*!μg/ml of ascorbic acid and 5*!mmol/L of beta glycerophosphate, the cells formed a mineralized extracellular matrix. Conclusion These results indicate that the cultured dental papilla cells keep their characteristics in vivo and the formation of extracellular mineralized tissue nodules in vitro provides a useful system to investigate calcification.
