摘要
建立了二氧化硅法纯化大熊猫粪便中DNA的技术 ,所得DNA可用于PCR扩增 ,而传统的有机溶剂提取的DNA不能去除粪便中抑制PCR反应的成分 ,使扩增结果呈假阴性。
This research aimed at setting up silica based DNA purification method to purify DNA from giant panda feces It is concluded that silica can bind bacterial genomic DNA at high concentration of KI and in neutral condition We use this condition to purify DNA by silica The result shows that we can purify bacterial genomic DNA by this method and purified DNA can be used to PCR reaction But classic organic solvent DNA purification method can not remove PCR inhibitors, which leads to false negative result
