T淋巴细胞Ag-NORs检测在肿瘤诊断中的意义

The significance of T lymphocyte nucleolar organizer region associated argyrophilic proteins examination in tumor diagnosis

目的通过检测健康人及良恶性肿瘤患者外周血 T 淋巴细胞核仁形成区嗜银蛋白(Ag～NORs),探讨其与机体免疫状况的关系,研究其在肿瘤诊断和病情监测中的应用价值。方法应用细胞银染技术对体外激活的人外周血 T 淋巴细胞染色,通过 KL 型肿瘤免疫图像分析系统检测Ag-NORs 含量(硝酸银染色酸性非组蛋白面积与细胞核面积的比值)即 IS%值。结果经统计学分析发现,健康人群中男女性的 T 淋巴细胞 Ag-NORs 的 IS%值分别为(7.77±0.61)%及(7.89±0.53)%,无显著性差异(P>0.05);良性肿瘤患者 IS%值为(7.66±0.60)%,比正常对照组(7.84±0.57)%略降低,但无显著性差异(P>0.05);恶性肿瘤患者 IS%值比正常对照组明显降低,为(4.46±0.69)%,健康人群及良性肿瘤患者与恶性肿瘤患者之间有显著性差异(P<0.05);在肺癌、胃癌、肝癌、大肠癌、乳腺癌等其他不同恶性肿瘤患者之间比较,结果分别为(4.46±0.75)%、(4.29±0.70)%、(4.62±0.69)%、(4.45±0.57)%、(4.50±0.69)%、(4.56±0.76)%,各种类型肿瘤患者间无显著性差异(P>0.05);恶性肿瘤患者不同临床时期即化疗前后 IS%值分别为(4.45±0.74)%、(3.86±0.71)%,有显著性差异(P<0.05);恶性肿瘤化疗后依据疗效不同(完全缓解、部分缓解、转移),IS%值随肿瘤病情恶化依次降低,分别为(6.57±0.47)%、(5.32±0.46)%、(3.47±0.42)%,有显著性差异。结论恶性肿瘤患者 T 细胞 Ag-NORs 值明显低于健康人群及良性肿瘤患者;外周血T淋巴细胞 Ag-NORs 检测在肿瘤诊断和病情监测等方面具有重要意义。

Objective To explore the relationship between T cell and immunity in cancer pa- tients by measuring the silver stain of nucleolus organizing regions(Ag-NORs).Therefore to find clues for the diagnosis and monitoring of cancer.Methods KL-2 imaging system was used to analyze the ar- eas of Ag-NORs in T cells.T cells isolated from peripheral blood were cultivated and stimulated,and silver staining was performed.Results The areas of Ag-NORs were analyzed.A total of 100 normal a- dults,35 patients with benign tumor and 112 patients with malignant tumor were observed.The aver- age IS% values were(7.84±0.57)%,(7.66±0.60)% and(4.46±0.69)% respectively.IS% was de- creased extremely in patients with malignant tumors compared with normal adults and patients with benign tumors.There were a significant difference between normal adults,tumors patients with benign tumors or malignant.No differences were observed between male (7.77±0.61)% and female (7.89± 0.53)% in normal adults.And-there were no differences among different kinds of cancers.Conclusion The value of Ag-NORs of T cell was significant lower in patients with malignant tumors and was sta- tistically different from that in normal adults and patients with benign tumors.The result demonstrated that the analysis of Ag-NORs might play an important role in the diagnosis and monitoring of cancer.
