摘要
目的探讨硒酸钠对军团菌生长的影响,为军团菌培养基的研制和改进提供依据。方法在含不同浓度硒酸钠的军团菌液体培养基中,接种一定浓度的军团菌悬液,分别于培养24 h、48 h、72 h移种于军团菌固体培养基,计算合不同硒酸钠浓度培养液增菌后军团菌生长的菌落数,用多因素方差分析的方法,分析硒酸钠对军团菌生长数量和生长速度的影响。结果在一定浓度范围内,硒酸钠对军团菌生长的促进作用呈浓度依赖关系,尤以0.025μg/ml硒酸钠对军团菌生长的促进作用最强, 当硒酸钠大于0.5μg/ml以上时,则明显抑制军团菌生长,且浓度越高,抑制作用越强;军团菌在合适量硒酸钠的培养基中孵育 24 h即可见菌落,而不合硒酸钠,相同培养须72 h才见菌落。结论在军团菌培养基中,加入0.025μg/ml硒酸钠能显著提高军团菌的生长数量,加快军团菌的生长速度。
Objective To study the effects of sodium selenate on Legionall growth, to provide the guideline for manufacture and ameliorate culture medium of Legionall.Methods Legionall were cocultured with sodium selenate in some different concentrations for 24 h, 48 h, 72 h, the subculturing to solid culture medium of Legionall respectively. The colony of Legionall in solid culture medium were examined at 72 h. All Dates were dealt by multifactor analysis of variance. Results The effect between sodium selenate and Legionall growth showed concentration-dependent, as 0.025 μg/ml of optimal level of sodium selenate make acceleration growth of Legionall. Legionall were inhibited when the concentration of sodium selenate more than 0.5 μg/ml. The Legionall colony can be observed at 24 h with appropriate concentration of sodium selenate in culture medium but 72 h without sodium selenate. Conclusion Legionall culture medium with 0.25 μg/ml sodium selenate can increase the quantity and improve the speed of Legionall growth.
