摘要
目的α-防御素抑制HepG2.2.15细胞HBV表达的体外初步探讨。方法不同浓度α-防御素于体外作用于HBV转染的HepG2.2.15细胞株模型,不同时间用免疫荧光法测定细胞培养上清液中HBsAg、HBeAg的量及HBV-DNA含量,并作出比较。结果作用24h后,α-防御素对HepG2.2.15细胞株HBV抑制更明显,且呈药物剂量依赖性。结论α-防御素在体外能有效地抑制HBV的表达,提示它在抗乙型肝炎病毒中有较大的应用前景。
Objective To investigate the inhibition of HBV expression on HepG2.2.15 cell line by α-defensin in vitro. Methods HepG2.2.15 clone cultured by DMEM was treated with the various concentration α-defensin,the HBsAg、HBeAg levels in surnatant were measurmented by immunofluorescent method and compared at different stage.Results The inhibition of HBV expression on HepG2.2.15 clone by α-defensin showed more efficient and was dose-dependent .Conclusion α-defensin can inhit the HBV expression effetively in vitro,which indicate it may be used as a antivities against HBV
