摘要
目的探讨伊文思蓝特异性染色技术快速鉴定新型隐球菌在临床检验医学诊断工作中的应用。方法配制伊文思蓝特异性染色液,评估伊文思蓝特异性染色液的稳定性及特异性;用伊文思蓝特异性染色液对新型隐球菌、白色念珠菌等常见真菌及白细胞染色,综合评价其染色效果,进一步比较伊文思蓝特异性染色液、阿利新蓝染色液、墨汁对新型隐球菌的染色效果。结果新型隐球菌被伊文思蓝特异性染色液染成蓝色或深蓝色,白细胞及其他常见真菌不着色,易于将新型隐球菌与白细胞及其他常见真菌区分;伊文思蓝特异性染色液稳定性好,染色效果稳定,优于阿利新蓝染色和墨汁染色,伊文思蓝特异性染色能在计数板中计数新型隐球菌,阿利新蓝染色易凝集对计数有一定的影响,而墨汁染色则无法计数。结论伊文思蓝特异性染色技术快速鉴定新型隐球菌设备要求简单、方法快速敏感,有很好的特异性和稳定性,适合各类医院推广应用。
Objective To study rapid identification of Cryptococcus neoformans by Evans blue specific staining technology in clinical laboratory diagnosis.Methods Prepare Evans blue specific staining reagent,to evaluate the stability and specificity of the reagent,using Evans blue specific staining reagent to staining fungus like Cryptococcus neoformans,Candida albicans and white blood cells,Comprehensively evaluating its staining effects,further comparing staining effects of Cryptococcus neoformans between Evans blue specific staining,and Alcian blue staining or ink staining.Results The Evans blue specific staining of Cryptococcus neoformans cells show the blue or deep blue appearance,but white blood cells and other fungus are not stained,it is very easy to differentiate Cryptococcus neoformans from white blood cells and other fungus.The stability of Evans blue specific staining reagent is very good,the Evans blue specific staining effect of Cryptococcus neoformans is better than Alcian blue staining and ink staining,Cryptococcus neoformans can be counted in counting plate under microscope by Evans blue specific staining and it can not be easy counted by Alcian blue staining because it is easy to agglutinate,also it can not be counted by ink staining.Conclusion Rapid identification of Cryptococcus neoformans by Evans blue specific staining technology need simple facilities.It’s fast and sensitive to detect,with good specificity and stability,and better for using in all hospitals.
