摘要
目的观察牛磺酸对创伤性颅脑损伤大鼠的脑保护作用,并初步探讨其机制。方法实验动物随机分为给药组、对照组和假手术组,通过皮层可控性打击器建立大鼠颅脑损伤模型。24 h后对大鼠进行神经功能缺损评分,测量脑组织含水量,采用酶联免疫吸附剂(enzyme linked immunosorbent assay,ELISA)测定脑组织丙二醛(malondialdehyde,MDA)含量、超氧化酶歧化酶(superoxide dismutase,SOD)活性。结果与对照组相比,给药组大鼠打击24 h后的神经功能缺损评分明显降低(P<0.05),脑含水量明显减少(P<0.05),脑组织MDA含量减少、SOD活性降低(P<0.05)。结论牛磺酸具有脑保护作用,其机制可能与减轻创伤后脑水肿的程度,减少MDA含量、降低SOD活性有关。
Objective To study the protective effect and possible mechanism of taurine on rat brain after traumatic brain injury.Methods The rats were randomly divided into taurine treatment group,control group and sham operation group.The models of rat brain injury were established by controlled cortical impact device,after 24 hours,evaluate the Neurological Severity Score and measure brain water content of rats,moreover,both the content of MDA and the activity of superoxide dismutase in brain tissue were measured by ELISA.Results Compared with the control group,the Neurological Severity Score of taurine treatment group significantly decreased( P < 0.05) and cerebral water content reduced obviously( P < 0.05),the content of MDA and activity of superoxide dismutase in brain tissue were reduced as well( P < 0.05).Conclusions Taurine has protective effect on brain,and the mechanism may be related to decrease the extent of brain edema,reduce the content of MDA and decrease the activity of SOD in brain tissue after trauma.
