摘要
目的 探讨用分子生物学方法检测血液Duffy血型基因型及该方法应用于HDN产前检查的可行性。方法 采用聚合酶链反应 限制性酶切片短长度多态性 (PCR RFLP)方法检测Duffy基因型。结果 用本研究采用的方法检出了 90例中国辽宁地区汉族血液样品Duffy血型的 3种基因型 ;检测出丈夫为Fy(a+b - )型、本人为Fy(a -b +)型的孕妇羊水样品 1例 ,其Duffy基因型为FYAFYB。结论 本方法可准确检测血液及羊水样品的Duffy基因型 。
Objective To evaluate a new molecular method for Duffy blood group genotyping and the possibility to apply this method to prenatal diagnosis of HDN.Methods PCR based restriction fragment length polymorphisms assay was designed to define Duffy blood group,in which the source of template DNA is derived from blood or amniotic fluid.Results Blood DNA samples from 90 unrelated Chinese individuals of Han population in Liaoning province,were analyzed with PCR RFLP and Duffy blood group was genotyped.3 genotypes were detected and the results of genotyping were consistent with those of serology;a DNA sample of amniotic fluid from a Fy(a-b+) pregnant woman,whose husband's Duffy phenotype was Fy(a+b-),was genotyped to be FYAFYB by PCR RFLP.Conclusion Blood and amniotic fluid samples can be correctly genotyped with PCR RFLP assay and the method an be applied to prenatal diagnosis of HDN resulting from anti Fya or anti Fyb.
