摘要 目的:研究与改良对流免疫实验试剂。方法:将破伤风梭菌经疱肉培养基培养后,培养液经离心、滤菌器过滤制备成未脱毒的破伤风梭菌外毒素。再将外毒素进行脱毒(化学法)和动物毒力试验,以获得无毒力的破伤风梭菌外毒素(粗制类毒素)。最后,以脱毒的破伤风梭菌外毒素作为抗原与TAT(抗体)进行对流免疫电泳实验。结果:改良后的试剂,其实验结果优于其它三种试剂。结论:改良后的试剂具有容易制备、低费用和安全性好的特点。 Objective: To research and improve CIEP experiment reagent. Methods: After the culture of tetanus clostridium in cooked meat culture medium, centrifugal sedimentation and bacteria filter, the tetanus clostridium exotoxin without detoxification is filtered. Then, the exotoxin detoxification (chemical method) and animal toxicity test are carried out to acquire avirulence exotoxin (rough out exotoxin). Finally, the detoxification clostridium tetani toxin is taken as antigen and TAT (antibody) is used to carry out CIEP experiment. Results: The experiment results of the improvemented reagents are better than that of the other three kinds of reagents. Conclusion: The improvemented experiment reagents have the characteristics of easy preparation and low cost and good security.
