乌灵胶囊对CBZ在海人酸致癫大鼠体内药代动力学的影响 被引量:2

摘要 目的研究乌灵胶囊对卡马西平(CBZ)在难治性癫痫(RE)大鼠体内药代动力学的影响。方法大鼠侧脑室注射海人酸(KA)诱导RE模型,将RE大鼠随机分为4组,分别灌胃给予生理盐水和乌灵胶囊(0.3、0.6、1.2 g/kg),连续灌胃给予7d后给予CBZ(80 mg/kg),于不同时间点采集血样,HPLC测定其中CBZ浓度,计算药动学参数。结果与单用CBZ组比较,联合给予中、高剂量乌灵胶囊可显著升高CBZ在RE大鼠体内的生物利用度。结论乌灵胶囊可影响CBZ在KA诱导癫痫大鼠体内的药动学过程,建议临床联合使用乌灵胶囊和CBZ治疗难治性癫痫时应当加强临床观察,必要时监测卡马西平血药浓度或降低CBZ的用量。 Objective To study the effect of Wuling Capsule on the pharmacokinetics of carbamazepine(CBZ)in refractory epilepsy(RE)rats.Methods The rat model of RE is induced by injection of kainate(KA)into the lateral ventricle of rats.The rats are randomly divided into 4 groups.After continuous administration of normal saline and Wuling capsules(0.3,0.6,1.2 g/kg)for 7 d,CBZ(80 mg/kg)is given.Blood samples are taken at time points,and the concentration of CBZ is determined by HPLC to calculate the pharmacokinetic parameters.Results Compared with the CBZ group alone,the combination of medium and high dose Wuling capsules significantly increased the bioavailability of CBZ in RE rats.Conclusion Wuling Capsule can affect the pharmacokinetics of CBZ in KA-induced epilepsy rats.It is suggested that clinical use of Wuling Capsule and CBZ in the treatment of refractory epilepsy should strengthen clinical observation and monitor the concentration of carbamazepine or if necessary.
