祖卡木颗粒和罂粟壳提取液对小鼠行为学的影响

摘要 目的探讨祖卡木颗粒(ZKM)和罂粟壳提取液(YSK)对小鼠行为学的影响。方法 ZKM2.7、5.4、10.8 g/kg,YSK 0.22、0.44、0.89 g生药/kg,连续给予昆明小鼠14 d,采用转棒试验观察祖卡木颗粒对小鼠运动、协调能力的影响;采用戊巴比妥钠催眠协同作用和自发活动试验,观察其对中枢神经系统的影响。结果与对照组比较,ZKM三组和YSK 0.22、0.44 g生药/kg组对小鼠睡眠持续时间无明显改变;给药组均对小鼠转棒试验动物潜伏期无明显改变;YSK 0.89 g生药/kg组小鼠睡眠持续时间明显降低(P <0.05),并可延长自主活动反应箱中心区域活动潜伏期(P <0.05);ZKM 10.8g/kg组自主活动反应箱四周活动持续时间明显降低(P <0.05)。结论临床等效剂量的祖卡木颗粒和罂粟壳提取液对小鼠运动、协调能力及中枢神经系统未见明显影响;高剂量祖卡木颗粒和罂粟壳提取液对小鼠自主活动有一定影响。 Objective To investigate the effect of super-clinical dose and course of treatment of Zukamu granules and poppy shell extract on behavior of mice.Methods Zukamu granules 2.7,5.4,10.8 g/kg;poppy shell extract 0.22,0.44,0.89 g crude drug/kg,Kunming mice are continuously given for 14 days,and the rod test is used to observe the movement of Zukamu granules on mice.The effect of coordination ability;synergy with sodium pentobarbital sodium and spontaneous activity test in mice to observe the possible potential effects of Zukamu particles on the central nervous system of mice.Results Compared with the control group,the Zukamu granule administration group and the poppy shell extract 0.22,0.44 g crude drug/kg group had no significant change in the sleep duration of the mice;the poppy shell extract 0.89 g crude drug/kg group mice continued to sleep.The time is significantly decreased(P<0.05);Zukamu granules and the poppy shell extract had no significant change in the incubation period of the mouse rotating rod test animals;the duration of activity around the autonomic activity reaction chamber of Zukamu granules 10.8 g/kg group is significantly reduced(P<0.05);0.89 g crude drug/kg group of poppy shell extract could prolong the activity latency of the central activity zone of the active activity reaction box(P<0.05).Conclusion Clinically equivalent doses of Zukamu granules and poppy shell extract had no significant effect on exercise,coordination and central nervous system in mice.But high-dose Zukamu granules and poppy shell extract(about 4 times the clinical dose)had an effect on the autonomic activity of mice.
