人皮肤组织褪黑素受体的鉴定及生物学意义 被引量:2

DETECTION AND LOCATION OF MELATONIN RECEPTOR IN HUMAN SKIN AND ITS MOLECULAR FEATURE

摘要 为证实褪黑素受体(melatoninreceptor,MR)存在于成人皮肤组织 ,并研究其亚细胞分布。应用RT PCR产物琼脂糖凝胶电泳及基因测序证实人皮肤组织存在MR ,并运用免疫组化法了解其亚细胞分布。PCR产物经琼脂糖凝胶电泳出现的阳性条带经纯化后基因测序的序列与GeneBank的mt1的cDNA序列相吻合 ,而未出现MT2 mRNA阳性条带 ;免疫组化证实人皮肤组织的细胞膜、细胞浆、细胞核均有MR亚型mt1蛋白的表达 ,未发现有MT2 蛋白的表达。 In order to detect and localize melatonin receptor and its mRNA expression in the human skin, human skin was obtained from healthy volunteers. Immunohistochemistry and RT-PCR were applied to detect MR in human skin and to identify its subcellular distribution. Electrophoresis of RT-PCR product showed positive band of mt 1 subtype in human skin. The subtype of MR, mt 1was present in the membrane, cytosol and nucleus of skin cells as shown by immunohistochemistry. It is suggested that skin is the target organ of melatonin.
