摘要
目的 :研究干扰素调节因子 7(Interferonregulatorfactor 7,IRF 7)对部分人肿瘤细胞凋亡的影响 ,探讨IRF 7抑制肿瘤生长的机制。方法 :将IRF 7的cDNA转染到人前列腺癌细胞 (TSU)和乳腺癌细胞 (MDA4 35 )中 ,采用Westernblot检测转基因细胞的caspase 3和Bax表达的变化 ;ELISA测定细胞裂解液中Bcl 2的含量 ,与转染空载体的对照组细胞进行比较。结果 :TSU和MDA4 35细胞转染IRF 7基因后 ,caspase 3和Bax的表达均增高 ,而Bcl 2的含量下降。结论 :干扰素调节因子 7(IRF 7)可能诱导肿瘤细胞出现凋亡 ,从而抑制肿瘤生长。
Objective:To study the effects of interferon regulator factor 7(IRF 7) on some human cancer cells apoptosis.Methods:Full length cDNA of IRF 7 was transfected into human breast cancer cell line(MDA435) and prostate cancer cell line(TSU).Caspase 3 and Bax from these transfectants were detected by Western blot comparing with control cells transfected with empty vector pcDNA3.Bcl 2 contents from lysates of transfectants were also determinted by ELISA method.Results:Expressions of caspase 3 and Bax from MDA435/IRF 7 and TSU/IRF 7 were much higher than that from control cells,whereas the Bcl 2 contents from transfectants were decreased.Conclusion:Interferon regulator factor 7(IRF 7) might have the ability to induce apoptosis in some human cancer cells in vitro.
