摘要
目的 :探讨阿魏酸哌嗪对糖尿病大鼠肾脏的保护作用及其机制。方法 :取 60只雄性SD大鼠 ,腹腔内注射链脲佐菌素 (60mg·kg- 1 )制造糖尿病模型。然后 ,将其中 30只大鼠给阿魏酸哌嗪液 1 0mg·d- 1 灌胃 ,共 8周。同时取 1 0只糖尿病大鼠和 1 0只健康大鼠作对照。实验前后分别观察各组大鼠血压、尿蛋白、肾功能、血浆血管紧张素Ⅱ (AngⅡ )含量和肾组织转化因子 (TGF β1 )和原癌基因 (c fos)的表达。结果 :阿魏酸哌嗪可控制糖尿病肾脏体积的扩大、血压的升高 ,减少尿蛋白的排出及改善肾功能 ,降低AngⅡ水平 ,同时还可减轻TGF β1 、c fos的表达。与对照组相比 ,差异均有显著性 (P <0 .0 5或P <0 .0 1 )。结论 :阿魏酸哌嗪可通过改善血液循环 ,降低血浆AngⅡ水平 ,控制血压 ,改善肾小球的“三高状态” ,从而抑制了ECM增生 ,达到保护肾功能 。
OBJECTIVE To investigate the protective effect and the mechanism of action of piperazine ferulate on experimental diabetic glomerulopathy.METHODS Experiments on rat diabetic glomerulppathy model induced by streptozotocin (60 mg·kg -1 ) were conducted. Changes of blood pressure, proteinuria, renal function (BUN, Cr), activity of angiotenisin Ⅱ expression of c fos and TGF β 1 were observed.RESULTS Compared with control group, the group administrated with piperazine ferulate presents significant lower levels of Scr, BUN,BP,proteinuria,AngⅡ( P < 0.01 ).It significantly decreases the expression of TGF β 1 and c fos gene in renal tissue ( P < 0.01 ).CONCLUSIONS Piperazine ferulate can improve the circulation, decrease the serum level of AngII and control blood pressure, so it suppresses the proliferation of ECM,prevents glomerulosclerosis and attenuates renal damage.
