摘要
目的研究漏芦乙醇提取物 (AEL)对正常大鼠及多种记忆障碍模型动物的学习记忆功能的改善作用 ,并初步探讨其作用机制。方法采用多种行为学方法考察AEL的益智作用 ,同时利用氧化震颤素致小鼠震颤及多种动物急性脑缺血实验探讨其作用机制。结果AEL能显著促进正常大鼠主动回避式条件反射的形成 ;明显改善戊巴比妥钠致小鼠记忆获得障碍、NaNO2 致小鼠记忆巩固障碍、东莨菪碱致小鼠空间辨别性障碍 ;增强氧化震颤素所致小鼠震颤的强度 ;剂量依赖性地延长急性脑缺血小鼠存活时间、降低急性脑缺血大鼠脑含水量。结论AEL具有促进学习记忆功能的作用 。
Objective To study the effects of the alcohol extract from Rhaponticum uniflorum(L.)DC.(AER)on the ability of learning and memory in normal rats and many amnestic animal models such as spatial discriminant disruption induced by scopolamine(SCPL).At the same time,the functionary mechanism was also studied.Methods The methods of the multiple training active avoidance response,step down tests and step through tests in mice were used to study the mentality improving effects of AER.At the same time,the methods,including tremor induced by oxotremorine in mice,acute cerebral ischemia in mice and acute cerebral ischemis in rats were used to investigate the functionary mechanism.Results AER can markedly facilitate the building of active avoidance response in rats,significantly improve the acquisition impairment of memory induced by pentobarbital sodium(PES),the consolidation disruption of memory induced by sodium nitrite(NaNO 2),the spatial discriminant disruption induced by scopolamine(SCPL)in mice.AER can enhance the strength of tremor induced by oxotremorine in mice,prolong the survival time of acute cerebral ischemia mice and reduce water content in the brains of the acute cerebral ischemia rats,which are dose dependent.Conclusions AER can improve the ability of learning and memory.Additionally,these effects might be related with its improving the function of the central cholinesterase systems and its protective effects of the cerebral cells from the damages of hypoxia and ischemia.
