摘要
在传统瑞替普酶的复性生产工艺中,采用了一种新型的生化工程的复性方法———PDI辅助复性,用50L、100L发酵罐进行中试,生产得率达到150mg/mL以上,比目前国外生产和国内科研中使用的谷胱甘肽还原法提高了3倍,是一个有希望在工业化生产中大大降低成本的新方法。
?Reteplase is an effective promising thrombolytics agent in clinics.Because reteplase was produced in the form of inclusion body,so we have to denature and dissolve the inclusion bodies,and renature the dissolved reteplase so as to obtain the correctly folded products.The recombinant human protein disulfide isomerase (rhPDI) was used to increased the reteplase.The yield was 150?mg/mL which was 3 times higher than the regular renaturation method of GSH∶GSSH.This novel method significantly reduced the cost of production.
