亚致死量放疗对Duchenne型肌营养不良鼠骨髓干细胞移植的影响 被引量:5

摘要 [目的]研究7 Gy γ射线放疗对Duchenne型肌营养不良鼠(mdx鼠)骨髓干细胞移植的效果.[方法]获取4~5周昆明鼠的骨髓干细胞,体外培养3 d,移植给7 Gy剂量放疗的7~8周的mdx鼠.22只7 Gy剂量放疗的肌营养不良鼠鼠,随机分为A、B、C、D 4组;A、B、C每组6只,静脉移植骨髓干细胞数为A组:1.2×106/只、B组4.8×106/只、C组1.2×107/只.D组4只作空白移植对照.观察受体鼠存活率,临床监测移植物抗宿主病(GVHD)情况;移植12周后,检测受体鼠肌组织抗肌萎缩蛋白表达情况.[结果]移植3个月后,22只实验鼠全部存活,18只移植鼠有近9%肌纤维表达dystrophin蛋白.[结论]移植前给予7 Gy γ射线放疗照射mdx鼠,部分破坏受体骨髓造血系统后,静脉注射移植同种、非同系鼠的骨髓干细胞,3个月之后,受照射鼠实现了100%存活;且移植鼠均有抗肌萎缩蛋白表达. [Objective] To study the effects of 7 Gy γ-ray irradiating dose on bone marrow stem cells transplantation for mice with Duchenne muscular dystrophy(mdx mice). [Methods] The bone marrow stem cells of Kunming mice were cultured in vitro for three days, then transplanted into mdx mice aged 7-8 weeks, which were pre-conditioned with 7 Gy γ-ray. 22 mdx mice , pre-conditioned by 7 Gy γ-ray, were devided into groups A, B, C and D randomly, with 6 mice in each group. The number of bone marrow stem cells transplanted was: 1. 2 ×106 in group A, 4. 8 ×106 in group B and 1. 2×107 in group C for each mouse . 4 mdx mice in group D served as the control group. The survival rate, GVHD reaction and dystrophin-expression of the transplanted mice after 12 weeks were observed. [Results] Three months after transplantation, all of the 22 mice were alive. Nearly 9% fibers of the muscle tissue of 18 transplanted mice expressed dystrophin protein . [Conclusion] Pre-conditioning with 7 Gy γ-ray and partly dystroying their bone marrow hematopoietic system , all of the irradiated mice were alive and all the transplanted mice expressed dystrophin protein.
