The Relationships Among the Expression of Vascular Endothelial Growth Factor, Microvessel Density and Radiosensitivity in Nasopharyngeal Carcinoma

摘要 [目的]探讨鼻咽癌组织中血管内皮生长因子(VEGF)表达及微血管(MVD)在预测鼻咽癌放射敏感性中的价值.[方法]对39例病理确诊为鼻咽癌的单纯放疗患者放疗前的鼻咽活检组织,进行Von Willebrand因子、VEGF免疫组化染色;用动态MRI观察肿瘤放疗后的体积变化,在MR机上直接测量肿瘤体积.[结果]VEGF表达与MVD密切相关(r=0.553,P<0.05).VEGF表达与肿瘤体积缩小百分比相关,有统计学意义(38~40 Gy时r=-0.45,P<0.05;68~76 Gy时r=-0.42,P<0.05).MVD与肿瘤体积缩小百分比的相关无统计学意义(38~40Gy时,r=-0.183,P=0.265;68~76 Gy时,r=-0.244,P=0.135).[结论]鼻咽癌组织内VEGF表达与放射敏感性呈负相关;VEGF表达与MVD呈正相关;MVD与放射敏感性的相关无统计学意义. [Objective] To evaluate the value of the expression of vascular endothelial growth factor (VEGF), microvessel density (MVD) in nasopharyngeal carcinoma in predicting the radiosensitivity (RS). [Methods] 39 patients, with their pathologic final diagnosis are nasopharygeal carcinoma, and only treated by radiotherapy were studied. Immunohistochemical analysis was performed on NPC biopsy specimen section. The sections were stained with polycolonal antibodies against VEGF and Von Willebrand factor. Dynamic MRI was employed to observe the regression of NPC after radiotherapy. The tumor volumes were measured on the MR console directly. [Results]There was a significant correlation between the expression of VEGF and MVD ( r = 0.553, P <0. 05). Significant correlation also existed between the expression of VEGF in NPC and the percentage of the volume of tumor regression(38 ~ 40 Gy, r=-0. 45, P < 0. 05, (68 -76) Gy, r = -0. 42, P < 0. 05 ) . There was no significant correlation between MVD and the percentage of the volume of tumor regression ( 38 ~ 40 Gy, r = - 0. 183, P = 0. 265, (68-76) Gy, r = -0. 244, P =0. 135) . [Conclusion] The expression of VEGF in NPC is negative correlated with RS. The expression of VEGF in NPC is positively correlated with MVD. There was no significant correlation between MVD in NPC and RS.
