摘要 [目的]探讨Fas相关磷酸酯酶(FAP-1)在结肠癌抵抗Fas介导的凋亡中的作用。[方法]通过免疫组化方法检测结肠癌组织中FAP-1、Fas受体(Fas-R)的表达及癌旁肿瘤浸润淋巴细胞Fas配基(Fas-L)的表达,通过TUNEL染色原位检测肿瘤细胞凋亡。[结果]56例结肠癌组织中,肿瘤细胞FAP-1阳性率为71%(40/56),FAP-1表达与组织分化程度及病理分期无关,P均>0.05。56例结肠癌组织中肿瘤细胞Fas-R免疫组化染色均阳性,且肿瘤组织中均可见到Fas-L免疫组化染色阳性的癌旁肿瘤浸润淋巴细胞浸润。FAP-1阴性的结肠癌组织中肿瘤细胞凋亡百分比较FAP-1阳性组高,(6.0±1.6)郴(4.0±1.2),P<0.05。FAP-1阴性的结肠癌组织中,肿瘤细胞凋亡与Fas-L阳性的肿瘤浸润淋巴细胞数目呈正相关,r=0.785,P<0.01;FAP-1阳性组未见此现象。[结论]结肠癌抵抗Fas介导的细胞凋亡与结肠癌细胞表达FAP-1有关。 [Objective] To evaluate whether Fas-associated phosphatase-1(FAP-1), an inhibitor of Fas signal transduction, is associated with resistance against Fas-R-mediated apoptosis in colon cancers. [Methods] FAP-1, Fas-Rand Fas-L were detected in colon cancer tissues using immunohistochem-istry. Apoptosis of cancer cells were detected in situ using terminal deoxynucleotidyltransferase mediated dUTP nick end labelling (TUNEL). [Results] FAP-1 expression was detected in 40 out of 56 resected colon adenocarcinomas(71% ) independent of the differentiation and staging of cancer ( P > 0. 05). Fas-R expression was observed in tumor cells of all colon adenocarcinomas tested. Fas-L immunostaining was observed in tumor infiltrating lymphocytes (TIL) adjacent to tumor nests. Percentage of apoptot-ic cancer cells was lower in colon cancers with positive FAP-1 immunostaining than in FAP-1 negative tumors , (4. 0 ±1. 2) vs( 6. 0 ±1. 6), P < 0.05. Positive correlation between the percentage of in situ apoptotic tumor cells and the number of Fas-L-positive tumor infiltrating lymphocytes was observed in FAP-1 negative colon cancers( r = 0. 785, P < 0. 01), but not in FAP-1 positive ones. [Conclusion] Resistance against Fas-mediated apoptosis in colon cancers is correlated with FAP-1 expression.
