摘要
目的 探讨阻断血小板源生长因子B链表达对裸鼠移植脑膜瘤的影响。方法 首先建立脑膜瘤裸鼠移植模型 ,观察转染PDGFB链反义寡核苷酸对移植瘤生长的影响。结果 PDGFB链反义寡核苷酸能够阻断裸鼠移植脑膜瘤表达PDGFB链 ,导致细胞增殖活性减弱 ,细胞生长受到抑制。
Objective To explore the feasibility of using PDGF B chain as a target for antisense gene therapy of meningioma in rats. Methods The meningioma nude mice model was established by implanting meningioma tissue into nude mice subcutaneously. An 18 mer antisense oligodeoxynucletide(AODN) complementary to a sequence starting at the ATG initiation codon of human c sis was synthesized and column purified. The nude mice bearing tumor were treated with PDGF B chain AODN and SODN by injecting transfection liquid into meningioma xenograft and their effect on the growth of meningioma xenograft were observed. Results Treatment with PDGF B chain AODN downregulated PDGF B B protein expression and inhibited the growth of meningioma xenograft. Conclusions PDGF B chain may be selected as target for gene therapy for meningiomas
