Expression and Its Significance of TIMP-2 Gene Protein in Gallbladder Carcinomas

目的 探讨TIMP 2蛋白在原发性胆囊癌中的表达及其与临床病理参数的关系。方法 研究包括 45例胆囊癌和 15例胆囊息肉样病变 ,相关临床病理资料如组织学类型、分化程度、临床分期、浸润深度、局部淋巴结转移及肝脏浸润等一并记录。应用免疫组化SABC方法结合图像分析技术定量检测上述病变的TIMP 2蛋白表达。结果 与胆囊息肉相比 ,胆囊癌TIMP 2表达虽无显著性差异 (P =0 .2 95 ) ,其表达与组织学类型和分化程度无关 ;但在不同Nevin分期、浸润深度、有否局部淋巴结转移及肝脏浸润的胆囊癌 ,其TIMP 2表达量存在显著差异。结论 TIMP 2表达能较准确地反映胆囊癌的生物学特点 ,可能对胆囊癌的临床分期。

Objective To study the relationships between expression of TIMP?2 gene protein and clinical-pathological parameters in gallbladder carcinomas.Methods Cases of Gallbladder carcinomas(n=45) and polypoid lesions of the gallbladder(n=15) were studied. Expression of TIMP?2 gene protein was examined by immunohistochemical avidin-biotin-complex method and image analysis.Clinical-pathological data such as histological type,grade of differentiation,level of infiltration,liver invasion and lymph node involvement,etc. were recorded.Results No significant difference in TIMP?2 expression was found between carcinoma and polypoid lesion of the gallbladder.Though significant correlation was not observed between TIMP?2 expression and histological type or differentiation degree, a significant relationship was found between TIMP?2 expression and different Nevin stage, infiltration level, local lymph node involvement or liver invasion (P<0.05). Conclusion Expression of TIMP?2 protein can reflect more accurately the biological character of gallbladder carcinomas. TIMP?2 might be acted as significant clinical indicators in the judgment of invasion or metastasis and the estimation of prognosis in gallbladder carcinomas.
