摘要
目的 :了解头孢哌酮对茶碱药代动力学 (药动学 )的影响 ,为临床合理使用茶碱提供数据。方法 :健康家兔 6只 ,受试前未使用过任何药物 ,静脉注射 4 .0 0mg·mL 1 茶碱溶液 1 0mg·kg 1 ,1 0min注射完毕。给药后于其耳缘静脉定时取血 ,采用高效液相色谱法测定家兔体内茶碱浓度 :用甲醇提取血清样品中的茶碱 ,色谱柱为Shim packODS C1 8;流动相为甲醇 :水 (70∶30 ) ,流速为 0 .8mL·min 1 ;检测波长为 2 52nm ;柱温为 30℃ ;进样量 1 0 μL。求算药动学参数。 结果 :头孢哌酮使茶碱的曲线下面积 (AUC) ,半衰期 (T1 2 )和表观分布容积 (Vd)下降 ;消除速率常数 (Ke)升高 ;清除率 (CL)无明显变化。结论 :头孢哌酮对茶碱药代动力学参数有影响 。
Objective:Probe the effect of cefoperazone on the pharmakinetics of theophylline so as to provide the data for using theophylline rationally in clinic. Methods:Six rabbits were given theophylline by venous injection at the dose of 10 mg·kg 1 . Blood samples taken from ear venous at different times were detected by HPLC. Draw the drug time curve of theophylline. After a week of taking cefoperazone at the dose of 40 mg·kg 1 i.m per day, draw the drug time curve again. to calculate the pharmacokinetics parameters. Results:Cefoperazone changed the parameter of theophylline as follows: AUC, T 1/2 and V d decreased; Ke increased while CL had no change. Conclusion:Cefoperazone has effect on the pharmacokinetics parameters of theophylline. The dose of theophylline should be modulated when combined with cefoperazone in clinical treatment.
