摘要
[目的]探讨甲状腺转录因子 (TTF_1)在甲状腺常见良恶性病变中的表达情况 ,并与甲状腺球蛋白(TG)和降钙素(CT)进行比较。[方法]采用Envision二步免疫组织化学分析法分别检测TTF 1和TG在45例常见的甲状腺良性病变和65例甲状腺癌中的表达 ,其中10例甲状腺髓样癌加做CT检测。[结果]TTF 1为核阳性 ,TG和CT为胞浆阳性。甲状腺滤泡细胞起源的良性病变TTF 1和TG阳性率均为100.0 % (45/45) ;而相应的恶性肿瘤TTF 1和TG的阳性率分别为89.1% (49/55)和85.5 % (47/55)。起源于甲状腺滤泡旁C细胞的髓样癌TTF 1阳性率为70.0 % (7/10) ,CT和TG阳性率分别为100.0 % (10/10)和0(0/10)。[结论]TTF 1在甲状腺常见良恶性病变中阳性率高 ,因其特异地表达在细胞核 ,是一种很好的甲状腺滤泡细胞和滤泡旁C细胞特异性标记物。
Purpose To investigate the expression of thyroid transcription factor_ 1( TTF-1) protein in benign and malignant thyroid diseases, and compared with those of thyroglobulin calcitoinin(TG)and(CT). TG and TTF-1 expression were detected by Envision technique in 45 specimens of benign thyroid diseases and 65 specimens of malignant thyroid neoplasms, and CT protein was also detected in 10 specimens of medullary carcinomas. TTF-1 were expressed in the nuclei of follicular cells and parafollicular C cells, whereas TG and CT were expressed in the cytoplasm of follicular and parafollicular C cells. TG and TTF-1 were both 100.0%(45/45) positive in follicular cells of benign thyroid diseases, and were 89.1%(49/55) and 85.5%(47/55) positive in malignant thyroid neoplasms respectively.In medullary carcinoma,positive rates of TTF-1, CT and TG were 70.0%(7/10), 100.0%(10/10)and 0(0/10) respectively. [Conclusion]TTF-1 were expressed in the majority of benign and malignant thyroid diseases. The immunostainning of TTF-1 was located in nuclear of cells.TTF-1 was a sensitive marker for thyroid follicular cells and parafollicular C cells.
