摘要 采用C18 色谱柱 ,体积比70∶30的甲醇 -0.02mol·L-1 磷酸二氢钾缓冲液 (含0.001mol·L-1 三乙胺 ,pH=6.60)作流动相 ,紫外检测波长230nm ,可待因作内标 ,建立了反相高效液相色谱法测定人血清中吗啡含量的方法 ;对流动相、样品预处理和储备液稳定性进行了考察 ,提出用乙酸乙酯直接萃取血清中的吗啡 ,所得样品稳定性好 ,萃取回收率为74 %~80 %;该法快速 ,适于生物样品中吗啡含量的测定。 A simple and rapid method for the determination of morphine in human serum was developed by RP- HPLC.Morphine in human serum was directly extracted with ethyl acetate without basification.Chromatographic separation of the extract was performed in Diamonsil C 18 column with an eluent consisting of methanol-phosphate buffer(70∶30by volume),and the eluate was monitored with UV detector at230nm.Codeine was used as an internal standard.Extraction of morphine in human serum using ethyl acetate has advantages of high extraction recoveries of74%~80%and good stability of the extract.The composition of eluent,pretreatment of sample and storage solvents for morphine were examined.
