摘要
采用热变性、聚乙二醇 (PEG) 60 0 0沉淀作用和DEAE 纤维素柱层析 3种方法对大蒜细胞溶质中的超氧化物歧化酶进行分离纯化。经过 3步分离纯化 ,从 5 0 0 g大蒜中得到15 6mg产品 ,比活为 314 5 5U/mg。酶的类型是Cu·Zn SOD ,在紫外区的最大吸收峰是 2 5 8nm。
In this research, SOD from Alliun sativum cell plasma was separated and purified by three methods,which is thermodenaturation, precipitation of polyethylene glycol(PEG) and the column chromatography of DEAE cellulose The product weighted 15 6?mg can be obtained from 500?g Allium sativum through three methods The specific activity of purified SOD is 3?145 5?U/mg This type of SOD is copper/zinc SOD The absorption apex exhibits at 258?nm
