摘要
目的 从人脑中克隆透明质酸结合蛋白(hbHABP)基因,研究其对各种肿瘤细胞体外增殖的影响。方法 根据已知透明质酸结合蛋白(HABP)DNA序列,设计引物,从人脑cDNA文库中克隆,构建真核细胞表达载体(pcDNA3-hbHABP),转染到人前列腺癌、乳腺癌、肺癌及黑色素瘤细胞中,观察体外细胞生长变化。结果 所有转染pcDNA3-hbHABP的肿瘤细胞相对于转染空载体pcDNA3的对照组细胞,出现生长受抑制现象,在软琼脂上的克隆形成数量也明显减少。结论 hbHABP可能是一种新的抑制肿瘤细胞生长的物质。
Objective To study the effects of human brain hyaluronan binding protein (hbHABP) on cancer cells proliferation. Methods hbHABP was amplified by PCR from the human brain cDNA database with 1 ,587 bp nucleotide, and the hbHABP was cloned into pcDNAS expression vector. hbHABP cDNA was thus transfected into human cancer cell lines as follows-. TSU (prostate cancer) , MDA435 (breast cancer) , Lewis lung (lung cancer) , and B16 (melanoma). Cell proliferation was studied by counting the cell numbers and the colonies on the soft agar plate. Results The growth of all transfectants was inhibited, and the ability of colony formation was reduced comparing with the mock transfectants. Conclusion The study suggested that hbHABP might present a new kind of anti - tumor substance in the human brain.
