病毒性心肌炎小鼠心肌细胞凋亡与外周血淋巴细胞凋亡的研究

The apoptosis of peripheral blood lymphocytes and myocytes in murine viral myocarditis

目的:研究心肌炎小鼠柯萨奇B3(CVB3)病毒性心肌炎模型外周血淋巴细胞凋亡与心肌细胞凋亡的关系。方法:采用AnnexinV/PI双参数法在流式细胞仪上定量检测CVB3心肌炎小鼠外周血淋巴细胞凋亡及心肌细胞凋亡百分率。结果:CVB3感染所致心肌炎小鼠外周血淋巴细胞凋亡以及心肌细胞凋亡均显著高于正常对照小鼠(P<0.001),且两者呈显著正相关(r=0.80,P<0.01);外周血淋巴细胞及心肌细胞凋亡百分率与心肌病理积分亦呈明显正相关(r=0.72,P<0.01;r=0.85,P<0.01)。结论:病毒性心肌炎外周血淋巴细胞凋亡可以反映心肌细胞凋亡情况,而且两者均与心肌炎病理损害程度密切相关。

Objective:To study the relationship between the apoptosis of peripheral blood lympho-cytes and myocytes in murine CoxsackievirusB 3 myocarditis.Methods :Annexin V/PI was used to detect quantita tively the percentage of apoptosis of lymphocytes and myocytes in CVB 3 mice with flow cytometry.Results:The percentage s of both lymphocyte apoptosis and myocyte apoptosis in murine myocarditis were sig nificantly higher than those in normal control mice(P<0.001),and there was a positive correlation between the two percentages(r=0.80).The two percentages were also positively correlated with the myocardial histopathological score(r=0.72and r=0.85,re spective ly).Conclusions :The percentage of apoptosis of lym-phocytes in peripheral blood could re flect the status of my ocyte apoptosis.The percentages of both lym pho-cyte apop tosis in peripheral blood and my ocyte apopto sis could be used to estimate the severity of my ocar-dial histopatholo gy.
