摘要 目的:研究脊髓源性神经干细胞的分离、培养、增殖和分化特点。方法:利用无血清培养和细胞克隆培养技术及免疫细胞化学方法,观察脊髓源性神经干细胞的体外扩增和贴壁分化情况。结果:脊髓源性神经干细胞具有增殖能力,并可传代培养,经含血清培养基培养,贴壁可分化为MAP2、GFAP和GC阳性细胞。结论:分离培养的脊髓源性神经干细胞具有增殖能力,并具有多向分化潜能。 Objective:To study the characteristics of isolation,culturing,multipotentiality and in-duced differentiation of neural stem cells derived from the spinal cord of embryonic rats.Methods:Serum free and clone culturing and im munocytochemistry were enlisted to observe the in vitro proliferation and differen tiation of the neu ral stem cells derived from the spinal cord and to identify differentiated neu ral cells.Results:The neural stem cells have the ability of proliferation and en dour passage.Nestin positive cells can be found in the daughter clones.In duced by10%serum,they can attach and differenti ate into MAP2,GFAP and GC positive cells.Conclusion:The neural stem cells the derived from the spinal cord of em bryonic rats have the ability of proliferation and multipotentiality.
