人脑胶质瘤中Bcl-2蛋白及Fas受体表达的研究 被引量:2

摘要 目的:探讨凋亡基因bcl-2与凋亡信号受体FasR在胶质瘤中的表达以及与胶质瘤的发生和治疗的关系。方法:采用流式细胞术方法检测8例正常脑组织及36例胶质瘤组织中Bcl-2蛋白及FasR的表达水平。结果:正常脑组织中无Bcl-2蛋白及FasR的阳性表达,而胶质瘤组织中Bcl-2蛋白的阳性表达率为38.89%(14/36),FasR的阳性表达率为75%(27/36)。与低度恶性胶质瘤(WHOⅠ-Ⅱ级)相比,高度恶性胶质瘤(WHOⅢ-Ⅳ级)中Bcl-2蛋白的阳性表达率明显升高(P<0.05)。结论:胶质瘤中高表达Bcl-2蛋白及FasR,提示Bcl-2基因过度表达与胶质瘤的发生有关,并为Fas配体或Fas单克隆抗体治疗胶质瘤提供参考。 Objection:To study the expression of Bcl-2protein and Fas receptor in human gliomas.Methods:The expression of Bcl-2protein and Fas receptor in36cases of glioma and8cases of normal brain tis sues was detected with flow cytometric method.Result:The expression level of Bcl-2pro tein and Fas re ceptor was negative in normal brain tissues,while positive ex pression of Bcl-2protein(defined as>10%)was observed in14cases of glioma (38.89%),and Fas receptor in27(75%).The expression level of Bcl-2protein were higher in high-grade glioma (WHO III-IV grade)than in low-grade gliomas(WHOⅠ-Ⅱgrade),P<0.05.Conclusion:The overexpression of Bcl-2gene and Fas recep tor in human glioma indi cates that Bcl-2gene may play an important role in the develop ment of malignant glioma.
