摘要
共聚焦显微镜是显微镜制作技术、光电技术、计算机技术相结合的产物 ,是现代化的光学显微镜 ,它对普通光镜做了多方面的技术改进 :用激光做光源并采用了共聚焦技术消除了透镜的色差和球差 ;运用点扫描技术 ,使标本上每一点的图像避免了相邻点的衍射光和散射光的干扰 ;结果用计算机处理 ;因而其图像高度清晰且数字化。激光共聚焦显微镜不仅可以观察活细胞、活组织的动态代谢过程 ,而且可以获得三维图像 ,与其他的生物学技术相配合 ,几乎可定性、定量定位地检测组织细胞内的任何一种生化成分。
Laser Scan Confocal Microscope (LSCM) is a modern optical microscope. It is a product that integrates microscope manufacturing technology, optical\|electronic technology, and computer technology. Compared with normal microscope, there are a lot of improvements. Because its light source is a laser tube, color aberration of lens and spherical aberration of lens were eliminated. The use of point scanning technology resulted in that, for every pixel of the image of the sample diffraction and dispersion of neighboring pixel was avoided. Results thus obtained were processed with computer, so the images were digital and very sharp. With the LSM, not only the dynamic metabolism process of live cells and tissues can be observed, but also three dimensional images can be obtained. Moreover, in combination with other biological technologies, almost every biochemical ingredient can be detected and measured in quantity and quality.
