摘要
目的 研究大鼠脑出血前脑内促凋亡基因半胱氨酰天冬氨酸特异性蛋白酶 - 3(caspase 3)的表达 ,以及三七总皂甙对其表达的影响。方法 用原位杂交法观察胶原酶诱导的脑出血大鼠前脑内caspase 3mRNA在给药组和对照组的表达变化 ;用免疫组化法观察caspase 3p2 0蛋白的变化。结果 脑出血后前脑内caspase 3mRNA及caspase 3p2 0主要在病灶及病灶周围、大脑皮质Ⅱ~Ⅴ层、海马CA2 、CA3区表达 ,阳性细胞包括变性神经元和胶质细胞。给药组阳性细胞数量显著减少 ,细胞着色变浅。结论 三七总皂甙可通过抑制caspase 3mRNA的转录及caspase 3蛋白的裂解活化 ,减少细胞凋亡 。
Objective To investigate the expression of a proapoptotic gene, cysteinyl aspartate specific proteinase 3 (caspase 3), in the forebrain and the effects of the total saponin from sanqi (the dried root of Panax notoginseng) on the expression of caspase 3 in the rats with intracerebral hemorrhage. Methods In situ hybridization was used to observe the changes in the expression of caspase 3 mRNA in the forebrain in the rats with intracerebral hemorrhage induced by collagenase in the total saponin group and the control group; immunohistochemical staining was used to observe the changes in the expression of caspase 3 p20. Results The expressions of caspase 3 mRNA and caspase 3 p20 mainly appeared in the area of the focus and around the focus, the cortical Ⅱ~Ⅴ layers and the hippocampal CA 2 - CA 3 subregions; and the positive cells appeared to be denatured neurons and gliocytes. In the animals in the total saponin group, there was a marked decrease in the number of the positive cells, the staining of which was pale in color. Conclusion The total saponin from sanqi can reduce apoptosis, and support the survival of the neurons and promote their repairing of damage in the forebrain in the rats with intracerebral hemorrhage, through inhibiting the transcription of caspase 3 mRNA and the activation of caspase 3.
