摘要
目的 :建立反相高效液相色谱法测定小鼠血浆中蒜氨酸浓度。方法 :用HPLC法 ,色谱柱为KromasilODS 1 (5μm ,4.6mm× 2 50mm) ,水为流动相 ,流速 0 .5mL·min- 1 ,紫外检测波长 2 2 0nm ,5—氟尿嘧啶为内标。结果 :蒜氨酸浓度与峰面积呈良好的线性关系 ,线性范围为 1 4 .8~ 1 4 81 .μg·mL- 1 。结论 :该法适用于蒜氨酸的药代动力学研究。结果表明 ,该方法经济、简便、快速。
Objective: To develop HPLC method for determination of alliin concentration in mice plasma. Methods: The plasma samples were extracted with methanol. The analysis involved a ODS 1 column as stationary phase and distilled water as mobile phase. The flow rate was 0.5mL·min -1 , UV detection wavelength was at 220nm. 5 Fu was used as the internal standard. Results: The calibration curve was linear over a range from 14.8μg·mL -1 to 148.1μg·mL -1 with a correlation coefficient of 0.9992. Conclusion: The method is economic, simple, sensitive and accurate.
