摘要
角朊细胞激活对T淋巴细胞增殖和细胞因子分泌的作用。TPA和IFN γ联合作用 ,激活角朊细胞表达表面分子 ,FACS检测MHCI、II类分子和CD80、CD86分子水平 ,RT PCR检测B7 H1共刺激分子表达。由激活角朊细胞提供T淋巴细胞激活的第二信号 ;绵羊红细胞花环沉淀分离纯化T细胞 ,抗CD3单抗提供第一信号 ,建立体外混合培养系统 ,3 H TdR渗入法检测T细胞增殖 ,ELISA检测培养上清细胞因子浓度。结果显示 ,TPA和IFN γ联合作用可诱导角朊细胞表达B7 H1共刺激分子 ,并上调其MHCII类分子。以B7 H1为第二信号 ,可以协同抗CD3单抗对T细胞的增殖作用 ,并呈现独特的细胞因子分泌格局。因此角朊细胞激活后表达B7 H1共刺激分子 ,此第二信号可刺激T淋巴细胞增殖 。
To investigate the role of activated keratinocytes on proliferation of T lymphocytes and secretion of cytokines.the keratinocytes were activated by joint treatment with TPA and IFN γ and the induction of surface molecule expression on keratinocytes were investigated. The expressions of MHC II molecules, CD80 and CD86 were assayed by FACS, and those of B7 H1 co stimulatory molecule detected by RT PCR. Employing the activated keratinocyte as the second signal and anti CD3 monoclonal antibody as the first signal, a mixed cell culture containing T lymphocytes purified by SRBC adsorption was established. The results showed that the co stimulatory molecule B7 H1 could be induced in keratinocytes by joint treatment with TPA and IFN γ. This co stimulatory signal enhanced the proliferation of lymphocytes cooperated with anti CD3 and presented unique cytokine production profile. It concludes that the activated keratinocytes express co stimulatory molecule B7 H1 on cell surface, and these molecules stimulate proliferation of T lymphocytes and regulate the functional differentiation of these cells.
