摘要
为了研究病毒性心肌炎 (VMC )心肌组织中MIP 1β和MCP 1的表达及其意义 ,以B3型柯萨奇病毒 (CVB3)腹腔注射BALB/c小鼠建立VMC小鼠模型 ,RT PCR实时定量分析正常对照组和VMC组心肌组织中CC家族趋化因子MIP 1β和MCP 1的表达 ,图像分析定量心肌组织切片浸润细胞数 ,以TCID50 分析心肌组织中CVB3载量变化 ,速率法检测血清CK MB水平。结果显示 :(1)MIP 1β和MCP 1在正常组均有表达 ,在VMC组MIP 1β和MCP 1的表达显著高于正常组 (P <0 0 1)。与 0天基础表达量相比 ,MIP 1β的表达在感染后第 4天达高峰 ,第 7、 9天下降。MCP 1在感染后第 4天上调表达 ,第 7、 9天时表达量持续升高 ,第 9天到高峰 ;(2 )在感染后第 4天时 ,心肌组织中开始出现单个核细胞浸润 ,7~ 9d浸润的单个核细胞逐渐增加。而正常心肌组织中未见明显的细胞浸润 ;(3)MCP 1的表达分别与浸润的单个核细胞数及CK MB水平相关 (均P >0 0 5 )、MIP 1β未呈明显相关。MIP 1β和MCP 1的表达与心肌组织中CVB3载量未呈明显相关性。说明MCP 1可能参与VMC心肌组织中单个核细胞的浸润。提示MCP 1可能与心肌组织中浸润细胞的效应功能发挥有关。
To investigate the expression of MIP 1β and MCP 1 and its implications in pathogenesis of viral myocarditis (VMC) induced by coxsackievirus group B type 3 (CVB3) BALB/c mice were inoculated intraperitoneally with CVB3 to establish animal model of VMC The expression of MCP 1 and MIP 1β (CC family) was detected quantitatively by real time RT PCR, while infiltrating cells were numbered by image analysis The level of CK MB was detected by rate method It was found: (1) The expression of MIP 1β and MCP 1 was detected both in VMC group and in normal control The expression of MIP 1β and MCP 1 increased significantly in VMC group vs in normal control ( P <0 01) The highest expression level of MIP 1β was found at 4 days in VMC and then the expression decreased continuously The expression of MCP 1 was climbedto a peak at 9 days (2)The infiltration of mononuclear cells in myocardium appeared at 4 days after CVB3 infection,while the number of infiltrating mononuclear cells increased at 7 days and 9 days The infittrotion was not found in myocardium of normal control. (3) The expression of MCP 1 was associated with the number of the infiltrating mononuclear cells and the level of CK MB( P <0 05), whereas there was not significant relationship with the expression of MIP 1β and the number of the infiltrating mononuclear cells and the level of CK MB The expression of MIP 1β and MCP 1 was not associated with CVB3 loading in myocardium These results demonstrated that MCP 1 may be involved in the infiltration of mononuclear cells of myocardium in VMC,and these also suggested that MCP 1 may be involved in the effector function of the mononuclear cells in the pathogenesis of VMC
