摘要
研究利用 2个混合引物的不同浓度梯度配比进行桑树RAPD扩增 ,得到明显不同的扩增图谱 ,随引物浓度的不同 ,扩增条带强弱、数目呈连续性变化 ,表明RAPD反应中引物混用具有可行性。引物浓度变化的实验结果再次证明RAPD反应中各反应物的竞争关系是形成扩增条带与理论推测存在差异的主要原因。当引物混用的配比为4 0 %~ 6 0 %时 ,结果较为稳定可靠。
Primer mixture is tested in this study.The profile of RAPD bands is successionally different from each other,both the bands number and intensity change with primer proportion varied in the same primers pair.The result showed that two primers mixture is a simple and powerful way to produce large number markers,which can increase the combining opportunity between template and primer.The result also showed that the competition between primer and template effects RAPD band profiles.The band profile is stable when two primers proportion varied between 40%~60%
