大鼠高血压性脑出血脑组织细胞凋亡表达及早期加压氧干预治疗对其影响 被引量:4

A STUDY OF APOPTOSIS EXPRESSION IN BRAIN TISSUE DUE TO HYPERTENSIVE CEREBRAL HEMORRHAGE IN RATS AND THE EFFECTS ON IT BY EARLY HYPERBARIC OXYGEN

摘要 ①目的 探讨大鼠高血压性脑出血双侧大脑皮质区和纹状体区细胞凋亡的时程变化和早期加压氧干预治疗对细胞凋亡的影响。②方法 将 15 6只Wistar雌性大鼠随机分为对照组、假手术组、高血压性脑出血4h~ 14d不同时间点自然恢复组和高血压性脑出血早期加压氧治疗后 6h~ 14d不同时间点恢复组。对照组为正常大鼠 ;高血压性脑出血动物模型采用双侧肾动脉前支部分热凝后给予高钠高脂高胆固醇饮食 6周 ,加小剂量胶原酶联合肝素立体定向尾壳核注入法建立 ;正常大鼠生理盐水代替胶原酶肝素尾壳核注入作为假手术组 ;早期加压氧治疗组在动物模型建立后 4h进行加压氧治疗。运用TUNEL法检测细胞凋亡数。③结果 自然恢复组4h~ 14d各时间点细胞凋亡数均明显高于对照组和假手术组 ,差异有显著性 (t=9.18~ 4 2 .5 6 ,P <0 .0 1) ;自然恢复组出血侧大脑皮质区和纹状体区各时间点 (14d除外 )的细胞凋亡数明显高于非出血侧 ,差异有显著性 (t =2 .71~ 18.39,P <0 .0 1) ,且其凋亡表达高峰在 18~ 2 4h出现 ,比非出血侧提前约 6h ;早期加压氧治疗组 12h~ 3d各时间点细胞凋亡数明显低于自然恢复组 ,差异有显著性 (t=2 .0 7~ 15 .34,P <0 .0 5、0 .0 1)。 Objective\ To investigate the changes of apoptosis expression with time in bilateral cerebral cortex and striatum of rats caused by hypertensive cerebral hemorrhage(HCH) and the influence of early treatment with hyperbaric oxygenation(ETHO) on apoptosis. \ Methods\ 156 mature female Wistar rats were randomly divided into normal control group, false operation group, natural recovery group and ETHO group. Establishment of hypertensive cerebral hemorrage: the anterior branches of bilateral renal arteries were partly coagulated, they were fed with high sodium and high fatty food for 6 weeks, then intracerebral hemorrhage was induced by injection of bacterial collagenase mixed with heparin into the caudate nucleus. In false operation group, normal saline instead of bacterial collagenase and heparin was injected into normal ratscaudate nucleus. In normal control group, the animals were normal rats. Hyperbaric oxygenation treatment was given at 4 h after HCH in ETHO group. The apoptotic cells in brain tissues were measured by TUNEL method. \ Results\ The apoptosis of natural recovery group, at 4 h-14 d ,were markedly more than that of both normal control group and false operation group, showing significant difference ( t=9.18-42.56,P <0.01). And the apoptotic cells on hemorrhagic hemisphere were markedly more than that of non hemorrhagic hemisphere, the difference was significant ( t=2.71-18.39,P <0.01). The apoptosis peak of the former appeared at 18-24 h, 6 hours earlier than the latter. The apoptotic cells of ETHO group greatly decreased than those of natural recovery group at 12 h-3 d , the difference was significant ( t=2.07-15.34,P <0.05, 0.01). \ Conclusion\ In rat HCH, the apoptosis peak of bilateral hemispheres appears at different time spot. All clinical treatments for HCH should be given 18 h before after the onset of HCH. ETHO is markedly effective to decrease apoptosis expression in HCH.
