用广西矮马制备抗人AFP血清

PREPARATION OF ANTI-AFP SERUM FROM GUANGXI DWARF HORSE

用亲和层析法提纯人甲胎蛋白(AFP)后免疫两匹广西矮马,制备马抗AFP血清取得成功。用双扩散法测定其抗AFP效价均在1∶32～1∶64以上。经免疫电泳鉴定:马抗AFP血清与正常人血清无沉淀线;与新生儿脐带血清及纯AFP溶液只有一条沉淀线。与放免法测定AFP含量在≥400μg/L的24份肝癌病人血清作对流免疫电泳测定,全部为阳性。和北京生物所891批AFP诊断血清作平行对流免疫电泳检测255份血样,其中43份同时阳性,212份同时阴性。单相特异性相一致。

Anti-AFP antibody was successfully prepared from two Guangxi dwarf horses immuno- lized with human AFP purified by immunoaffinity chromatography.The titers of the antibodies were higher than 1:32～1:64 determined by double immuno-diffusion.The results of immuno-electrophoresis showed that such horse anti-AFP antibody did not produce any precipitate line,with normal human serum,but with ambilical cord serum and purified AFP solution it only produced one preciptate line.The antibodies reaction were all positive in the counter immunoelectrophoresis assay in which anti-serum(AFP ≥400μg/L)from 24 hepatocellular carcinoma patients were used as the antigens.When 255 samples of serum were assayed by parallel counter immuno-electrophoresis,in which batch No.891 AFP diagnosis serum manufactured by Peking Biological Products Institute was used as the control anti-sermun,43 of the samples were both positive and 212 of them were both negative,indicating that their specibilities were of the same order.
