优质HMW谷蛋白亚基转基因小麦的获得及其遗传稳定性和品质性状分析 被引量:42

摘要 利用编码除草剂PPT抗性的bar基因作为选择标记,构建了含有优质HMW谷蛋白亚基1Dx5和IDy10基因的植物表达载体pBPC30,用基因枪导入京花1号、京411号和京冬6号等受体品种的幼胚和幼穗,获得了一批转基因小麦植株(T0)及其后代株系(T1~T5).PCR,点杂交和Southern杂交鉴定表明,外源基因已整合到转化植株T0的基因组,并稳定表达.转基因植株T1代能抗20~80 mg/L的ppT,其中1株抗性超过150 mg/L.经SDS-PAGE分析表明,外源IDx5和IDy10基因在T2代不同株系中获得了表达.转基因系TG3-74缺失了亚基8,但亚基2,5,10和12都表达,同时出现了一个新的亚基位于亚基7与原有亚基8之间(暂记为8).品质分析结果表明,有5个株系的Zeleny沉淀值为44.2~49.0 mL,与对照CNN(48.0 mL)相当,分别比京花1号(26.9 mL)和京冬6号(30.7 mL)提高了59.6%~64.3%.T5代株系的粉质曲线稳定时间比受体品种(3~7 min)大幅增长,转基因株系TG5-23b的稳定时间为16 min,rGl-28d、TG3-76b、TG5-98和TG5-99的面团稳定时间高达30min以上.
