摘要 目的 探讨异氟醚对大鼠不同脑区NMDA受体 (NR)的功能亚单位NR1磷酸化的影响 ,揭示吸入麻醉药抑制NR开放的机理。方法 SD大鼠 2 0只 ,随机分为对照组 (吸氧 )和实验组(吸异氟醚 ) ,Western blot方法检测吸入异氟醚后大鼠大脑皮层、脑干、海马的磷酸化NR1蛋白量以及NR1蛋白量的变化。结果 大鼠吸入 1 75 %异氟醚 2 0min后。大脑皮层的磷酸化NR1蛋白量比对照组下降 4 1 6 % ,脑干、海马的磷酸化NR1蛋白量也分别比对照组下降 4 9 8%和 39 9% (P <0 0 1) ;但NR1蛋白表达量未发生显著改变 (P >0 0 5 )。结论 异氟醚可抑制NR1的磷酸化 ,这种作用是吸入麻醉药抑制NR通道开放的重要原因 ;而NR1细胞内C Objective To investigate the mechanisms,by which inhalational anesthetics lead to the inhibition of NR activity,we examined the effects of isoflurane on phosphorylation of NR1 subunits.Methods Twenty SD rats were randomly divided into control group(inhalation of oxygen)and experimental group(inhalation of isoflurane).Using Western blot technique,we compared the changes in phosphorylated NR1 protein and NR1 protein expression in the cerebral cortex,cerebellum and hippocampus of two groups.Results After inhalation of 1 75% isoflurane,phosphorylated NR1 protein was markedly decreased by 41 6%,49 8% and 39 9%( P< 0 01) in the cerebral cortex,cerebellum and hippocampus tissue respectively.No significant changes of NR1 protein expression were observed in these tissues.Conclusions Isoflurane can decrease phosphorylation of NR1 subunit,which are likely to take part in the mechanisms,by which isoflurane leads to the inhibition of NR activity.C terminal phosphorylation site of NR1 subunit may be the direct or indirect site of isoflurane action.
