基因枪法导入Bar基因转化春小麦幼胚的研究——影响转化的主要因素

Studies on Bar gene transformation of immature embryos in spring wheat (Triticum aestivum L.) by particle bombardment --The important factors effected on transformation

利用基因枪法将抗除草剂 Bar基因导入春小麦东农 774 2 ,IE4 7和辽 10。在本试验条件下以培养 2 0～ 2 2 d的幼胚为受体 ,在 80 ︼g·枪金粉量和 6 cm的轰击距离下的基因枪转化效果最佳。

Plasmid pBARGUS,containing selectable Bar gene and GUS reporter gene,was directly delivered into immature embryos in three cultivars of spring wheat (NE7742,IE 47 and Liao 10)by particle bombardment.The results showed that the best transformation of immature embryos was obtained with embryogenic callus induced in vitro around 20～22 days and by particle bombardment with 80 μg of gold particles and 6 cm distance per bombardment in the studies.$$$$
