摘要
建立高效的植株再生体系是遗传转化的重要基础。以北方主栽水稻品种为试材 ,将 Thidiazuron(简称TDZ)应用于水稻愈伤组织的不定芽分化研究 ,探讨了 TDZ等植物生长调节剂对水稻愈伤组织不定芽分化和生长的影响。结果表明 ,TDZ显著促进水稻愈伤组织的不定芽分化 ,其细胞分裂素活性均大大高于 KT和 6 - BA,最佳使用浓度仅为 KT和 6 - BA的 1/ 10~ 1/ 2 0。不同 TDZ浓度对不定芽的分化效率在不同品种上有差异 ,东农 4 2 1品种在浓度为 0 .10 m g· L- 1时诱导效率最高 ,平均芽数达到 7.7个 /块 ,诱导效率达到 5 5 8.30 % ;东农 92 - 11品种在浓度为 0 .2 0 m g· L- 1时效果最佳 ,平均芽数为 7.9个 /块 ,分化效率高达 5 5 3%。高浓度的 TDZ有一定的毒害作用 ,导致愈伤组织褐化 ,诱导的不定芽出现矮化、叶片卷曲。
The establishment of effective regeneration system is a very important basis of genetic transformation.Selecting the main rice varieties in north area,the paper focuses on the influence of TDZ on the differentiation of adventitious buds in th aim of improving the percentage of plant regeneration from callus.The results show that:TDZ can significantly accelerate the differentiation of adventitious buds.TDZ has a higher activity that 6 BA and KT with about 1/10~1/20 of the activity of them.The effect of TDZ concentration on the differentiation of adventitious buds varies from different varieties.As TDZ is 0.1 mg·L -1 ,the rate of differentiation of Dongnong 421 can reach the highest value of 539%,and mean buds can reached 7.7/ explant.As TDZ is 0.20 mg·L -1 ,the rate of differentiation of Dongnong 92-11 can reach the highest value of 553%,and mean buds can reached 7.9/explant.However,high concentration of TDZ is poisonous to callus of rice.If concentration of TDZ is higher,callus of rice would be browning,dwarfness wrinkled leaves,and vitrification.
