摘要
目的 :藻酸钙凝珠复合骨髓基质细胞经体外培养扩增后行细胞活力、组织形态学和电镜超微结构观察 ,探讨其应用于组织工程软骨修复缺损的可能性。方法 :传代培养的骨髓基质细胞 ,复合藻酸钙制成凝珠 ,细胞密度为 1× 1 0 6 /ml,体外培养 4周后 ,行倒置相差显微镜、台盼蓝染色、甲苯胺蓝染色、透射电镜检查。结果 :骨髓基质细胞培养后大量扩增 ,与藻酸钙复合良好并能在其中保持活性及分裂能力。结论 :藻酸钙复合骨髓基质细胞用于组织工程方法修复软骨缺损 ,具有较强的可行性。
Objective:To observe bone marrow stromal cells vitality,histomorphology and ultrastructure in vitro culture combined with calcium alginate beads.To try to find out the possibility of this compound material to articular cartilage tissue engineering.Methods Bone marrow stromal cells from the New Zealand white rabbits were harvested,expanded in cell culture,mixed with calcium alginate beads in a cell density of 1×10 6/ml,in vitro culture 4 weeks,then observed with trypan blue stain,toluidine blue stain and transmission electron microscopy.Results:After in vitro culture,the cells and the material compound well developed,bone marrow stromal cells had activity and reproduce capability.Conclusion:Calcium alginate-bone marrow stromal cells composite may succeed in constructing engineered articular cartilage.
