人类端粒酶基因反义核酸对恶性肿瘤细胞端粒酶抑制的实验研究 被引量:1

摘要 目的 :研究人类端粒酶 (hTR)基因的反义核酸对K5 62细胞、Hep 2细胞端粒酶活性和K5 62细胞凋亡的影响。方法 :采用TRAP法检测K5 62细胞、Hep 2细胞在反义核酸处理前后端粒酶活性的变化以及采用TUNEL法观察反义核酸处理前后K5 62细胞凋亡的变化。结果 :hTR基因反义核酸能有效抑制K5 62细胞和Hep 2细胞的端粒酶活性 ,并且诱导K5 62细胞发生凋亡。结论 :hTR基因反义核酸能显著抑制肿瘤细胞的端粒酶活性 。 Objective: To study the inhibitory effect of human telomerase activity in K562 and Hep 2 cells and the apoptosis by an antisense oligonucleotide against human telomerase RNA(hTR) in K562 cells.Methods: Telomerase activity of K562 and Hep 2 cells with antisense treatment was measured using the TRAP assay and apoptosis was studied by means of TUNEL method.Results: The telomerase activity was effectively inhibited and the apoptosis was induced by the antisense treatment.Conclusion: Antisense oligonucleotide against hTR can effectively inhibit the telomerase activity of tumor cells,and subject is an effective and novel approach for the treatment of malignant cancers.
