黄芪注射液预处理对兔未成熟心肌保护作用的研究

Experimental Studies of Astralus Injection Preconditioning on Cardioprotective Effect in Immature Rabbit Heart

目的 探讨单纯黄芪预处理对兔未成熟心肌再灌注 (I/ R)损伤的影响。方法 2 4只幼兔 (14～ 2 1d)随机分 3组 ,每组 8只 ,利用 L angendorff模型灌注其离体心脏。平稳灌注 3 0 m in后 ,向球囊缓慢注入生理盐水 ,调整至左室舒张末压 (L VEDP)为 10 mm Hg(1mm Hg=0 .13 3 k Pa)。对照组继续灌注 15 min;黄芪预处理组 :则用黄芪注射液灌注 15 min。两组心脏均经历停灌 3 0 min(保温、保湿 )、缺血自动停跳及再灌注 40 min复制全心 I/ R模型。观察各组的血液动力学、冠脉流出液心肌酶、心肌能量变化、病理学变化及分子生物学改变。结果 黄芪预处理组左心功能、冠脉流量恢复及再灌后心肌组织内 ATP含量明显优于对照组 ,心肌酶值明显低于对照组 ,心肌细胞线粒体半定量分析显示预处理组线粒体损伤轻于对照组 ,心肌诱导型一氧化氮合酶 i NOS高于对照组。

OBJECTIVE The purpose of this study was to investigate whether immature rabbit hearts could be protected by Astralus Injection preconditioning. METHODS Twenty four healthy white rabbits of 3 4 weeks old ,weighing 300 350 g were randomly divided into three groups: control group (n=8),IPC group (n=8), Astralus injection(20mg/ml)group (n=8). In IPC group the hearts were first subjected to an IPC stimulus consisting of 5 minutes of global ischemia plus 10 minutes of reperfusion.In AI group the heart were perfused by AI (20 mg/ml) 15 minutes. The hearts in three groups were subjected to 30 minutes of global ischemia and 45 minutes of reperfusion. Krebs Henseleit buffer bubbled with a 95%O2 5%CO2 gas mixture perfused the hearts in langendorff mode at 70 cmH 2O. Perfusate temperature and the heart temperature when undergoing ischemia were both maintained at 38℃. During and after reperfusion, the change of myocardial hemodynamics, myocardial enzyme in the coronary effluent, myocardial energetic metabolism, morphology and iNOS were determined. Ultrastructual changes in the mitochondria were evaluated semiquantitatively. RESULTS LVDP, ±dp/dtmax, CF recovery had a significant improvement in AI group. LDH, CK levels were the least in AI group, and AI group also had a higher level of tissue ATP and a higher level of tissue iNOS. CONCLUSIONS AI preconditioning can protect immature rabbit hearts.
